CJC-1295 / Ipamorelin

What it is

CJC-1295 is a long-acting analog of growth-hormone releasing hormone (GHRH), and ipamorelin is a selective growth-hormone secretagogue that mimics ghrelin’s action at the GHSR-1a receptor. Studied together, the pairing is intended to combine two distinct upstream signals in the growth-hormone axis: CJC-1295 extends the duration of GHRH stimulation, while ipamorelin was described in its original pharmacology paper as achieving growth-hormone release without meaningfully elevating cortisol or ACTH (Raun et al., Eur J Endocrinol, 1998, PMID 9849822). What the literature has explored

In a healthy-adult dose-escalation study, single subcutaneous doses of CJC-1295 produced dose-dependent increases in growth hormone lasting six or more days and increases in IGF-1 lasting nine to eleven days, with an estimated half-life of roughly six to eight days (Teichman et al., J Clin Endocrinol Metab, 2006). A companion trial found that growth-hormone pulsatility was preserved during continuous CJC-1295 stimulation, meaning trough and mean GH rose without flattening the body’s normal episodic release pattern (Ionescu et al., J Clin Endocrinol Metab, 2006, PMID 17018654).

Ipamorelin’s foundational pharmacology work in animal models described a selective GH-releasing effect without the cortisol or prolactin elevation seen with some older secretagogues, which is part of why it is frequently paired with GHRH analogs rather than used to replace them (Raun et al., 1998, PMID 9849822). It is worth noting that a Phase II CJC-1295 trial in a specific patient population was terminated, and the broader secretagogue class still lacks large, long-term human trials for body-composition or wellness use (Transl Androl Urol review, PMC7108996).
